The potential molecularly therapy target to <i>MYCN</i>‐amplificated neuroblastoma

نویسندگان

چکیده

Abstract Neuroblastoma (NB) is an embryonal deadly cancer in childhood driven by MYC or MYCN‐driven oncogenic signaling. The amplification of MYCN leads to malignant progression NB and poor prognosis. Traditional chemotherapy still a standard treatment for NB. Most the cytostatic drugs take function anti‐neuroblastoma. However, children with exhibit different clinical outcome variability biological characteristics compared adult patients. Given this, it urgent need explore novel, safer more efficacious treatments.

The Canonical Wnt Signaling (Wnt/β-Catenin Pathway): A Potential Target for Cancer Prevention and Therapy

Precise regulation of signal transduction pathways is crucial for normal animal development and for maintaining cellular and tissue homeostasis in adults. The Wnt/Frizzled-mediated signaling includes canonical and non-canonical signal transduction pathways. Upregulation or downregulation of the canonical Wnt-signaling (or the Wnt/β-Catenin signal transduction) leads to a variety of human diseas...

Harnessing the potential of epigenetic therapy to target solid tumors.

Epigenetic therapies may play a prominent role in the future management of solid tumors. This possibility is based on the clinical efficacy of existing drugs in treating defined hematopoietic neoplasms, paired with promising new data from preclinical and clinical studies that examined these agents in solid tumors. We suggest that current drugs may represent a targeted therapeutic approach for r...

WIP1 Phosphatase as a Potential Therapeutic Target in Neuroblastoma

The wild-type p53-induced phosphatase 1 (WIP1) is a serine/threonine phosphatase that negatively regulates multiple proteins involved in DNA damage response including p53, CHK2, Histone H2AX, and ATM, and it has been shown to be overexpressed or amplified in human cancers including breast and ovarian cancers. We examined WIP1 mRNA levels across multiple tumor types and found the highest levels ...

Erk5 Is a Potential Therapeutic Target in Alk-positive Neuroblastoma

Anaplastic lymphoma kinase (ALK) has been implicated as an oncogenic driver in pediatric neuroblastoma and is frequently activated by amplifi cation and gain-of-function mutations. However, results from phase I trials have suggested that, in contrast to other tumor types such as non–small cell lung cancer, single-agent therapy with the ALK inhibitor crizotinib is not effective in pediatric pati...

PPM1D is a potential target for 17q gain in neuroblastoma.

Neuroblastomas (NBs) show complex patterns of genetic abnormalities, which may include amplification of the MYCN gene, deletion of 1p, or a gain of DNA at 17q, the last being the most frequent observation in NB tumors. However, the specific genes and the molecular mechanisms responsible for development and progression of NB remain poorly understood. We investigated aberrations of DNA copy numbe...
